if you don't tell her parents, and something terrible happened to your niece, you'd carry that guilt forever.

I would encourage her parents to find a way around this knowledge, ie, find an untruth in something he says, so that the niece never knows you gave up the confidence in you (lest next time they don't find out). but, myself, i'd have to tell this information (just like schools and hospitals having to report suspected abuse). Somehow this situation has to be addressed.
